The thesis project aims to evaluate which pharmaceutical products are available on the market and which ones are still being tested, for the treatment of glaucoma. In the paper, after an ocular anatomy and pathology’s description, the traditional pharmaceutical forms, such as solutions and suspensions, which are 90% of commercially available anti-glaucoma preparations, are taken into consideration. Then the strategies to improve therapeutic efficacy that have been adopted in the last years in order to overcome these formulations’ limits when they are applied on the ocular surface, are discussed. Moreover innovative pharmaceutical forms (most of them not yet approved by Food and Drug Administration) are described, such as eye inserts, implants, contact lenses and colloidal systems. They have been designed and developed to increase anti-glaucoma therapy efficacy and to improve patients compliance thanks to a drug sustained release.
Il progetto di tesi ha come obiettivo quello di valutare quali prodotti farmaceutici sono disponibili sul mercato e quali ancora in fase di sperimentazione per il trattamento del glaucoma. Dopo una dettagliata descrizione dell’anatomia oculare e della patologia in esame, sono prese in considerazione le forme farmaceutiche anti-glaucoma più diffuse sul mercato, rappresentate per il 90% da soluzioni e sospensioni. Sono quindi discusse le strategie formulative adottate negli ultimi anni per superare le limitazioni di queste formulazioni quando applicate sulla superficie oculare al fine di migliorarne l’efficacia terapeutica. Sono, inoltre, descritte forme farmaceutiche innovative (per la maggior parte ancora non autorizzate dalla Food and Drug Administration), tra cui inserti oculari, impianti, lenti a contatto e dispersioni colloidali, progettate e sviluppate al fine di aumentare l’efficacia della terapia anti-glaucoma e migliorare la compliance dei pazienti, garantendo un rilascio prolungato del farmaco veicolato
