Crohn's disease is defined as a chronic nonspecific pathology of the whole digestive system, belonging to the IBD. Crohn's disease has several variable aspects; this heterogeneity can be linked to the location of the lesions, the behavior of the disease and the presence of local or systemic complications. Signs and symptoms can change considerably from person to person. Generally observed in patients: inflammation, chronic intermittent diarrhea, recurring abdominal pain, partial or complete obstruction. The main cause of Crohn's disease is still unknown, and you think that is to be searched for in a combination of several factors, including immunological, genetic, infectious and environmental factors. Not being still known the cause of the disease, there isn't a decisive pharmaceutical treatment. In this thesis, after an overview about the disease and conventional therapeutic approaches, the attention has focused on a new treatment option which includes the use of nanotechnology. The nanotechnology allows targeted delivery and controlled drug. The hopeful data obtained experimentally are waiting for clinical trials; Soon, patients with Crohn's disease will be treated with medications conveyed only where they are needed, thanks to the continued rise of nanotechnology.
La malattia di Crohn viene definita come una patologia cronica aspecifica dell'intero apparato digerente, appartenente alle malattie infiammatorie croniche intestinali. Il morbo di Crohn ha molti aspetti variabili; questa eterogeneità può essere legata alla localizzazione delle lesioni, al comportamento della malattia e alla presenza di complicanze locali o sistemiche. I segni e i sintomi possono variare notevolmente da persona a persona. Comunemente nei pazienti si osservano: infiammazione, diarrea cronica intermittente, dolore addominale ricorrente, ostruzione parziale o completa. La causa fondamentale del morbo di Crohn è ancora oggi sconosciuta, e si pensa che sia da ricercare in una combinazione di diversi fattori, tra i quali fattori immunologici, genetici, infettivi e ambientali. Non essendo ad oggi nota la causa della malattia, non esiste un trattamento farmacologico risolutivo. In questo lavoro di tesi inoltre, dopo un quadro generale della patologia e degli approcci terapeutici convenzionali, è stata focalizzata l'attenzione su una nuova opzione terapeutica comprendente l' uso delle nanotecnologie. L'impiego delle nanotecnologie permette un rilascio mirato e controllato dei farmaci. I promettenti dati ottenuti sperimentalmente sono in attesa di studi clinici; presto i pazienti con malattia di Crohn potranno essere trattati con farmaci veicolati solo dove sono necessari, grazie alla continua ascesa delle nanotecnologie.
