The purpose of this thesis is the study of two validations on a peculiar matrix: Canadian poplar wood chips. The validations to be carried out concern pesticides (102 analytes) and mycotoxins (ochratoxin A). The project was the result of a collaboration between the University of Pavia and LabAnalysis in the master’s degree plus (LM+) project. The methods used have already been validated by LabAnalysis and this thesis aims to extending these methods to the matrix under consideration. Pesticides were extracted with acetone and purified with gel permeation chromatography. The analysis was then carried out in UPLC-MS/MS and GC-MS/MS. Ochratoxin A was first extracted in acetonitrile and water, followed by purification on immunoaffinity columns. The resulting extract was analyzed with UPLC-MS/MS. The study finally focused on the determination of the linearity, specificity and repeatability of the method, along with its intermediate precision and quantification limit.
Questo lavoro di tesi magistrale riguarda due validazioni su una matrice al quanto peculiare: trucioli di legno di pioppo canadese. Le convalide da effettuare riguardano i pesticidi (102 analiti) e le micotossine (ocratossina A). Il progetto nasce dalla collaborazione dell’Università di Pavia con LabAnalysis attraverso il progetto di Laurea Magistrale Plus (LM+). I metodi applicati sono già stati validati da LabAnalysis e questa tesi è stata finalizzata ad estendere tali metodi per la matrice in esame. Per i pesticidi è stata eseguita un’estrazione in acetone, una purificazione in gel permeation chromatography e successivamente l’analisi è stata condotta in UPLC-MS/MS e GC-MS/MS. Per l’ocratossina A è stata eseguita un’estrazione con acetonitrile e acqua, una purificazione su colonnine immunoaffini e successivamente l’analisi è stata condotta in UPLC-MS/MS. Per la validazione del metodo sono state studiate e verificate: la linearità, la specificità, la precisione, la ripetibilità, la precisione intermedia e il limite di quantificazione.
