Inflammatory bowel disease (IBD) comprises a group of chronic inflammatory disorders, including Crohn's Disease (CD) and Ulcerative Colitis (UC), which mainly involve the gastrointestinal tract but can also present extraintestinal manifestations. The main shared features between the two diseases are symptom-based, while they differ significantly in terms of the cytokines released and the immune landscape. The role of different cytokines has been widely studied during the last decades but the role of the cytokine eNAMPT in emergency haematopoiesis and myelopoiesis remains poorly understood. This study aims to investigate its role in both steady-state and emergency conditions related to inflammatory bowel disease, characterizing its impact on mobilization and expansion of HSPCs in the bone marrow. Moreover, a scRNA-seq analysis has been performed to identify a transcriptional signature during acute and chronic experimental colitis. Together, these findings identified eNAMPT as a potential therapeutic target in the dysfunction of hematopoietic cell production associated with IBD.
Le malattie infiammatorie croniche intestinali (IBD, Inflammatory Bowel Disease) comprendono un gruppo di disturbi infiammatori cronici, tra cui il morbo di Crohn (CD, Crohn's Disease) e la colite ulcerosa (UC, Ulcerative Colitis), che coinvolgono principalmente il tratto gastrointestinale ma possono anche presentare sintomi extraintestinali. Le principali caratteristiche condivise tra le due patologie sono di natura sintomatologica, mentre esse differiscono significativamente per quanto riguarda le citochine rilasciate e il quadro immunitario. Il ruolo delle diverse citochine è stato ampiamente studiato negli ultimi decenni, ma il ruolo della citochina eNAMPT nell'ematopoiesi di emergenza e nella mielopoiesi resta oggi ancora poco chiaro. Questo studio si propone di indagare il suo ruolo sia in condizioni basali (steady-state) sia in condizioni di emergenza legate alle IBD, caratterizzandone l'impatto sulla mobilizzazione e sull'espansione delle cellule staminali e progenitrici ematopoietiche (HSPCs) nel midollo osseo. Inoltre, è stata condotta un'analisi di scRNA-seq per identificare i cambiamenti a livello trascrizionale durante la colite sperimentale acuta e cronica. Nel loro insieme, questi risultati identificano eNAMPT come un potenziale bersaglio terapeutico nella disfunzione della produzione di cellule ematopoietiche associata alle IBD.
