Glioblastoma (GB) IDH-wildtype is the most aggressive primary brain tumor, characterized by a striking metabolic plasticity, which promotes chemoresistance. This thesis work investigates the early bioenergetic adaptation mechanisms induced in U251 human glioblastoma cells by exposure to sublethal concentrations (equal to the 〖"IC" 〗_25) of two innovative platinum complexes, "Pt(IV)Ac-POA" and 〖"Pt(acac)" 〗_2 "(DMS)" , in comparison with the standard drugs Cisplatin and Temozolomide (TMZ). The investigation was conducted by coupling Western Blot quantification with immunofluorescence morphological analysis of the markers GLUT1, GLUT3, "ATP5A" , and "HIF-1" α. The results highlight drug-specific responses: while the global protein level of GLUT1 remains stable, all treatments induce its delocalization from the plasma membrane to the cytoplasm. The expression of the high-affinity transporter GLUT3 is drastically reduced by Cisplatin and 〖"Pt(acac)" 〗_2 "(DMS)" , whereas TMZ and "Pt(IV)Ac-POA" alter its compartmentalization, dispersing the characteristic perinuclear clusters. At the mitochondrial level, the "ATP5A" subunit shows a quantitative trend toward an increase, which is accompanied, however, by a severe punctate fragmentation of the organelle network under the effect of Cisplatin, TMZ, and "Pt(IV)Ac-POA" . Finally, under normoxic conditions, TMZ and "Pt(IV)Ac-POA" stabilize "HIF-1" α; however, the alkylating agent drives a massive nuclear translocation, whereas the platinum(IV) complex maintains a diffuse cytoplasmic signal. In conclusion, while 〖"Pt(acac)" 〗_2 "(DMS)" exhibits a predominantly glycolytic-suppressive action similar to Cisplatino, the "Pt(IV)Ac-POA" prodrug displays a multi-level efficacy that disorganizes glucose import systems and mitochondrial architecture, effectively disrupting the cell's compensatory survival programs.
Il Glioblastoma (GB) IDH-wildtype è il tumore cerebrale primitivo più aggressivo, caratterizzato da una spiccata plasticità metabolica che ne favorisce la chemioresistenza. Questo lavoro di tesi analizza i meccanismi precoci di adattamento bioenergetico indotti sulle cellule U251 dall'esposizione a dosi subletali (〖"IC" 〗_25) di due complessi innovativi al platino, "Pt(IV)Ac-POA" e 〖"Pt(acac)" 〗_2 "(DMS)" , in confronto ai farmaci standard Cisplatino e Temozolomide (TMZ). L'indagine è stata condotta accoppiando la quantificazione tramite Western Blot all'analisi morfologica in immunofluorescenza dei marker GLUT1, GLUT3, "ATP5A" e "HIF-1" α. I risultati evidenziano risposte farmaco-specifiche: mentre il livello globale di GLUT1 resta stabile, in tutti i trattamenti si osserva una sua delocalizzazione dalla membrana plasmatica al citoplasma. L'espressione del trasportatore ad alta affinità GLUT3 viene drasticamente ridotta da Cisplatino e 〖"Pt(acac)" 〗_2 "(DMS)" , mentre TMZ e "Pt(IV)Ac-POA" ne alterano la compartimentazione, disperdendo i caratteristici cluster perinucleari. A livello mitocondriale, la subunità "ATP5A" mostra un trend quantitativo all'aumento, a cui si associa però una severa frammentazione puntiforme della rete d'organello sotto l'effetto di Cisplatino, TMZ e "Pt(IV)Ac-POA" . Infine, in condizioni di normossia, TMZ e "Pt(IV)Ac-POA" stabilizzano "HIF-1" α; tuttavia, l'alchilante ne guida una massiva traslocazione nucleare, mentre il complesso al platino(IV) mantiene il segnale citoplasmatico. In conclusione, sebbene 〖"Pt(acac)" 〗_2 "(DMS)" mostri un'azione prettamente glicolitico-soppressiva simile al Cisplatino, il profarmaco "Pt(IV)Ac-POA" esibisce un'efficacia multilivello che disorganizza i sistemi di importazione del glucosio e la struttura mitocondriale, andando a inibire efficacemente i programmi compensatori di sopravvivenza.
