I G-quadruplexes (G4s) are non-canonical secondary structures of DNA and RNA involved in the regulation of numerous cellular processes and considered potential therapeutic targets. In this context, naphthalene diimides (NDIs) represent a class of molecules of particular interest due to their ability to interact with G4 structures, while ruthenium complexes exhibit physicochemical and biological properties that make them promising for antitumor applications. The present work was therefore focused on the design and synthesis of NDI–Ru(III) hybrid molecular systems, with the aim of combining the G4 recognition properties of the NDI platform with the characteristics of the metal center. To this end, the NDI core was functionalized through nucleophilic aromatic substitution reactions, optimizing the synthetic conditions for the introduction of different side groups and subsequently developing conjugation strategies with the Ru(III) complex. Different coupling approaches were investigated, and the obtained molecules were characterized using appropriate analytical techniques. The synthesized systems constitute a molecular platform for further studies aimed at evaluating their G-quadruplex recognition properties and their potential interest in the antitumor field.
I G-quadruplex (G4) sono strutture secondarie non canoniche del DNA e dell’RNA, coinvolte nella regolazione di numerosi processi cellulari e considerate potenziali bersagli terapeutici. In questo contesto, le naftalendiimmidi (NDI) rappresentano una classe di molecole di particolare interesse per la loro capacità di interagire con le strutture G4, mentre i complessi di rutenio mostrano caratteristiche chimico-fisiche e biologiche che li rendono promettenti per applicazioni antitumorali. Il presente lavoro è stato pertanto dedicato alla progettazione e sintesi di sistemi molecolari ibridi NDI–Ru(III), con l’obiettivo di combinare le proprietà di riconoscimento dei G4 della piattaforma NDI con le caratteristiche del centro metallico. A tale scopo, il core NDI è stato funzionalizzato mediante reazioni di sostituzione nucleofila aromatica, ottimizzando le condizioni sintetiche per l’introduzione di differenti gruppi laterali e successivamente sviluppando strategie di coniugazione con il complesso di Ru(III). Sono state investigate diverse modalità di accoppiamento e le molecole ottenute sono state caratterizzate mediante le opportune tecniche analitiche. I sistemi sintetizzati costituiscono una piattaforma molecolare per successive valutazioni volte a studiarne le proprietà di riconoscimento dei G-quadruplex e il potenziale interesse in ambito antitumorale
