Poly-γ-glutamic acid (γ-PGA) is an extracellular polymer produced by Bacillus subtilis with diverse biological and biotechnological applications. The γ-PGA biosynthetic operon, pgs, is activated by phosphorylated DegU (DegU-P) and SwrA. The DegS-DegU two-component system is a well-characterized regulatory pathway that controls a wide range of physiological processes occurring during the stationary phase of growth in B. subtilis. Conversely, much less is known about the small regulatory protein SwrA, despite its established roles in bacterial motility and γ-PGA production, both mediated by its interaction with DegU-P. γ-PGA production is strongly influenced by environmental conditions, including temperature, but the regulatory mechanisms underlying its temperature dependence remain obscure. This study investigates the molecular mechanisms underlying temperature-dependence of γ-PGA biosynthesis in B. subtilis, addressing the contributions of SwrA and DegU-P. It was verified that pgsB expression, which reflects the activation of the γ-PGA biosynthetic pathway, was substantially reduced at 24 °C compared with 37 °C. Analysis of swrA mRNA secondary structure predictions identified a potential hairpin structure that could sequester the ribosome-binding site at lower temperatures, functioning as a potential RNA thermometer. To test this hypothesis, a targeted mutation was introduced to disrupt the predicted structure and its potential role in temperature-dependent swrA expression was investigated. It was shown that the mutation did not restore γ-PGA production at 24 °C. Furthermore, the wild-type swrA allele showed higher expression than the mutant swrA allele at both 24 °C and 37 °C. Consistent with this observation, swrA overexpression from an inducible promoter did not restore γ-PGA production at 24 °C. In addition, aprE expression, known to be induced by DegU-P alone and on which SwrA has a negative impact, was investigated as a readout of DegU-P activity. In a ΔswrA background, aprE expression was strongly reduced at 24 °C compared with 37 °C. Together, these findings indicate that the temperature dependence of γ-PGA production is primarily associated with reduced DegU-P activity at 24 °C rather than with temperature-dependent swrA expression. The mechanism that controls the temperature-dependence of DegU-P activity remains to be investigated.

L'operone biosintetico del γ-PGA, pgs, è attivato dalla forma fosforilata di DegU (DegU-P) e da SwrA. Il sistema a due componenti DegS-DegU è un sistema di regolazione ben caratterizzato che controlla un'ampia gamma di processi fisiologici che avvengono durante la fase stazionaria della crescita in B. subtilis.
