FLVCR1a and FLVCR1b are two isoforms encoded by the FLVCR1 gene, contributing to cellular heme homeostasis; however, their molecular and functional properties remain incompletely defined, in particular the mitochondrial isoform FLVCR1b. Here we characterized both FLVCR1a and FLVCR1b with particular emphasis on FLVCR1b. We optimized a purification strategy for FLVCR1b that yields material suitable for biochemical, functional, and further structural analyses. Immunofluorescence confirmed the mitochondrial localization of FLVCR1b, consistent with its role in mitochondrial heme trafficking. Moreover, we provide convergent evidence that FLVCR1b forms a dimer under native conditions, using size exclusion chromatography (SEC), native PAGE, pull-down assay, and native mass spectrometry (nMS). Comparative Fab and ligand screening revealed distinct interaction profiles for FLVCR1a and FLVCR1b, indicating isoform-specific three-dimensional and conformational properties despite their common genetic origin. Functional transport assays with liposome-reconstituted FLVCR1a and FLVCR1b showed that heme and related porphyrins modulate transport activity in a substrate-depending manner. Finally, computational structural analysis identified candidate residues potentially involved in heme recognition and binding in FLVCR1a as well as in FLVCR1b, and corresponding site-directed mutants were generated for future experimental validation.
FLVCR1a e FLVCR1b sono due isoforme codificate dal gene FLVCR1, che contribuiscono all’omeostasi cellulare dell’eme; tuttavia, le loro proprietà molecolari e funzionali non sono ancora state completamente definite, in particolare per quanto riguarda l’isoforma mitocondriale FLVCR1b. In questo studio abbiamo caratterizzato sia FLVCR1a che FLVCR1b, ponendo particolare enfasi su FLVCR1b. Abbiamo ottimizzato una strategia di purificazione per FLVCR1b che consente di ottenere materiale idoneo per analisi biochimiche, funzionali e ulteriori analisi strutturali. L’immunofluorescenza ha confermato la localizzazione mitocondriale di FLVCR1b, in linea con il suo ruolo nel traffico mitocondriale dell’eme. Inoltre, forniamo prove convergenti del fatto che FLVCR1b formi un dimero in condizioni native, utilizzando la cromatografia a esclusione dimensionale (SEC), PAGE nativo, saggio di pull-down e spettrometria di massa nativa (nMS). Lo screening comparativo di Fab e ligandi ha rivelato profili di interazione distinti per FLVCR1a e FLVCR1b, indicando proprietà tridimensionali e conformazionali specifiche per ciascuna isoforma nonostante la loro comune origine genetica. I test funzionali di trasporto con FLVCR1a e FLVCR1b ricostituiti in liposomi hanno dimostrato che l’eme e altre porfirine modulano l’attività di trasporto in modo dipendente dal substrato. Infine, l’analisi strutturale computazionale ha identificato residui candidati potenzialmente coinvolti nel riconoscimento e nel legame dell’eme sia in FLVCR1a che in FLVCR1b, e sono stati generati corrispondenti mutanti sito-specifici per una futura validazione sperimentale.
