Alzheimer’s (AD) and Parkinson’s (PD) diseases are two of the most studied pathologies in the recent years, since they are currently urgent and important unmet medical needs. The underlying mechanisms of these diseases are not yet fully understood. In addition there are few approved therapies for them, for that, the research about new biochemical and chemical entities able to cope with these pathologies is fundamental. The enzyme fat mass and obesity associated protein (FTO), a m6A demethylase, is involved in various pathological processes, including AD and PD. Recently, it has been reported that vitamin E succinate can interact with FTO and its natural E3 ligase, DTX2, thereby promoting FTO degradation. Therefore, derivatives of vitamin E similar to vitamin E succinate may function as both FTO inhibitors and degraders. The latter function could be achieved by using these derivatives as part of PROTACs. In this work we report the development and optimization of a new synthetic pathway to get access to a small library of new vitamin E derivatives.
