The endosomal trafficking system plays a fundamental role in regulating plasma membrane proteins and sorting them for recycling or degradation. In plants, ubiquitinated membrane proteins are recognised by the ESCRT machinery and delivered to the vacuole for degradation. Although the ESCRT pathway is evolutionarily conserved in animals, fungi and plants, the ESCRT-0 complex is absent in higher plants. Instead, TOL (Target of Myb1) proteins act as ubiquitin receptors during the early stages of this pathway. While Arabidopsis thaliana contains nine TOL proteins with partially redundant functions, the liverwort Marchantia polymorpha has only two homologues, MpTOLa and MpTOLb. This reduced genetic redundancy, together with its predominantly haploid life cycle, makes M. polymorpha a useful model for investigating TOL function. MpTOLa is more closely related to AtTOL1/AtTOL2, whereas MpTOLb resembles AtTOL6/AtTOL9. This study aimed to investigate the roles of MpTOLa and MpTOLb by analysing mutant phenotypes. CRISPR/Cas9 vectors containing two guide RNAs per locus were used to introduce mutations into the target genes. Stable mutations maintained after vegetative propagation were identified in two Mptola lines (Mptola-1 and Mptola-2) and three Mptolb lines (Mptolb-5, Mptolb-6 and Mptolb-8). Sequence analysis predicted early frameshift mutations in the Mptola lines, potentially causing loss of function, and mutations leading to C-terminal truncations in the Mptolb lines, retaining the N-terminal core. Their effects on transcript and protein stability and function remain to be experimentally verified. Phenotypic differences were observed between mutant lines and their corresponding wild-type (WT) controls. Both Mptola lines showed reduced size and asymmetric thallus development, including an approximately 90° rotation of one lobe. Mptola-1 also displayed altered rhizoid growth and curled rhizoid tips, whereas Mptola-2 exhibited more vertical, hyponastic growth. Mptolb-5 developed wider lobes, asymmetric thallus growth and an earlier-forming apical notch. Mptolb-6 showed reduced growth and a compact, strongly folded thallus with a pronounced three-dimensional structure. In Mptolb-8, the apical notch was more pronounced than in the WT, and differences in thallus expansion became more evident during later development. Gemma cup formation also differed between mutant and WT plants. The male mutants (Mptola-1 and Mptolb-6) developed gemma cups later than the male WT control Tak-1, whereas the female mutants (Mptola-2, Mptolb-5 and Mptolb-8) developed them earlier than the female WT control Tak-2. These observations suggest that MpTOLa and MpTOLb may contribute to cell polarity, organogenesis and developmental timing in M. polymorpha. Based on known TOL functions in Arabidopsis, altered trafficking of proteins involved in auxin signalling may contribute to the growth phenotypes, while changes in ABA-related trafficking may be associated with differences in gemma cup formation. Both hypotheses require further experimental investigation. A second aim was to generate molecular tools for future complementation experiments using AtTOL1, AtTOL2, AtTOL6 and AtTOL9 to complement M. polymorpha TOL mutants and investigate protein subcellular localisation. Two expression vectors, MpEF1ap::AtTOL2-GFP and MpEF1ap::AtTOL9-GFP, carrying Arabidopsis TOL genes fused to a C-terminal GFP tag under the constitutive MpEF1ap promoter, were successfully assembled. Completing the remaining constructs (MpEF1ap::AtTOL1-GFP, MpEF1ap::AtTOL6-GFP, MpEF1ap::MpTOLa-GFP and MpEF1ap::MpTOLb-GFP) would enable broader comparisons of TOL protein localisation and complementation capacity. Overall, this study provides tools to investigate MpTOLa and MpTOLb functions in plant development and the conservation of TOL-mediated sorting.
