Fanconi anemia (FA) is a rare hereditary bone marrow failure syndrome caused by mutations in genes crucial for DNA interstrand cross-link (ICL) repair and replication fork stability. Although the activation of the FA/BRCA pathway is widely characterized at the protein level, its response coordination at the transcriptional level and the regulatory involvement of long non‑coding RNAs (lncRNAs) remain poorly defined. This study aimed to characterize the global transcriptional remodeling induced by genotoxic stress in HeLa cells, focusing on the temporal dynamics of FA/BRCA pathway genes and lncRNAs, and to investigate the upstream regulatory networks mediated by the E2F1 and E2F4 transcription factors.

Abstract
Fanconi anemia (FA) is a rare hereditary bone marrow failure syndrome caused by mutations in genes crucial for DNA interstrand cross-link (ICL) repair and replication fork stability. Although the activation of the FA/BRCA pathway is widely characterized at the protein level, its response coordination at the transcriptional level and the regulatory involvement of long non‑coding RNAs (lncRNAs) remain poorly defined. This study aimed to characterize the global transcriptional remodeling induced by genotoxic stress in HeLa cells, focusing on the temporal dynamics of FA/BRCA pathway genes and lncRNAs, and to investigate the upstream regulatory networks mediated by the E2F1 and E2F4 transcription factors.| File | Dimensione | Formato | |
