This thesis investigates the potential neuroprotective role and effects of an in vivo treatment with an experimental compound (EC) on striatal medium spiny neurons (MSNs) in the R6/2 mouse model of Huntington’s Disease (HD). Using ex vivo whole-cell patch-clamp recordings, the study evaluates intrinsic membrane properties, basal synaptic transmission and corticostriatal synaptic plasticity (LTP). Results indicate that while the EC does not reverse intrinsic membrane hyperexcitability or active firing properties, it seems to restore the frequency of spontaneous excitatory postsynaptic currents (sEPSCs) and rescue corticostriatal LTP. These findings demonstrate that the compound acts as a network-level modulator, protecting corticostriatal synapses from mutant huntingtin toxicity.

Abstract
These findings demonstrate that the compound acts as a network-level modulator, protecting corticostriatal synapses from mutant huntingtin toxicity.| File | Dimensione | Formato | |
